刺孢小克银汉霉氨基酰化酶拆分DL-丙氨酸的研究 总被引:1,自引:0,他引:1
选育了刺孢小克银汉霉(Cunninghamellaechinulata)9980菌株,并对其进行液体培养,比较了3种不同培养基中菌体细胞氨基酰化酶活性,考察了几种因素对菌体细胞拆分反应的影响,并对DL-丙氨酸进行了光学拆分。结果表明:蛋白胨培养基菌体细胞酶活最高,达680u/g。菌体细胞拆分反应最适温度55℃,最适pH7.0,最佳底物浓度为0.2mol·L-1,缓冲体系中的无机离子对拆分反应有抑制作用,10-3~10-4mol·L-1的Co2+对拆分反应有激活作用。用菌体细胞对DL-丙氨酸拆分反应中,D-丙氨酸得率平均达87.1%。 相似文献

For the first time, a nodulin-like gene promoter was isolated from Gossypium hirsutum L. Guo Y18 by means of inverse PCR. Three plant expression vectors were constructed for functional identification of the promoter. These vectors were different only in promoter regions; three truncations of the nodulinlike promoter took the place of the CaMV35S promoter in the pBI121 plant expression vector. Then, the three vectors were introduced into cotton plants via the pollen tube pathway. The expression patterns of the gus gene driven by nodulin-like promoter truncations were investigated in the offspring of transgenic cotton plants. Histochemical GUS staining and fluorescence quantitative analysis were performed to achieve this goal. The results showed that the nodulin-like promoter was a strong, highly reproductive organspecific promoter, which demonstrated a much higher driver activity than the CaMV35S promoter did in cotton reproductive organs, but relatively lower activity in vegetation. Identification of the speciality and strength-determining regions of the nodulin-like promoter was also undertaken. 相似文献

本研究旨在探讨并比较慢性间歇性低压低氧(intermitten thypobaric hypoxia,IHH)和慢性连续性低压低氧(continuous hypobaric hypoxia,CHH)对大鼠血液动力学作用的影响。40只成年Sprague—Dawley大鼠随机分为5组:对照组(CON),28天IHH处理组(IHH28),42天IHH处理组(IHH42),28天CHH组(CHH28)和42天CHH组(CHH42)。IHH火鼠于低压氧舱分别接受28或42天模拟5000m海拔高度低氧(11.1%O2)处理、每天6h。CHH处理大鼠生活在低压氧舱环境中,除每天半小时常氧供食、供水和清洁外,其余时间均分别接受时程为28或42天的模拟5000m海拔高度低氧(11.1%O2)处理。每周定时测定大鼠体重。通过导管法测定基础常氧和急性低氧状态下的血液动力学,包括半均动脉压(meanartery blood pressure,MAP)、心率(heartrate,HR)、左审收缩峰压(1eft ventricular systolic pressure,LVSP)、正负左率最人压力变化速率(maximum change rate of left ventricular pressure,±LVdP/dtmax)。通过生物化学方法测定大鼠心肌超氧化物岐化酶活性和丙二醛含量。并分别测定全心、左心室和右心室重量。结果显示:(1)CHH42大鼠基础HR和MAP低于CON,IHH和CHH28大鼠(P〈0.05)。(2)IHH大鼠表现出明显的抗心肌缺氧/复氧损伤作用,表现为急性低氧状态下的HR、MAP、LVSP和+LVdP/dtmax,改变明显低于CON大鼠(P〈0.05);CHH大鼠表现出更为明显的抗急性低氧心脏保护作用,表现为急性低氧的HR、MAP、LVSP和±LVdP/dtmax;改变明显低于CON和IHH火鼠(P〈0.05),但出现复氧损伤作用,表现为复氧过程中血液动力学的恢复明显低于CON和IHH大鼠(P〈0.05)。(3)与CON大鼠相比较,IHH和CHH大鼠心肌抗氧化能力明显增强(P〈0.05,P〈0.01)。(4)与IHH和CON大鼠相比较,CHH大鼠表现明显的右心室肥厚(P〈0.01)。结果表明,IHH可诱导有效的心脏保护作用,而无明显的不良反应,因而具有潜在的实际应用价值。 相似文献

为研究中国汉族群体IDUA 基因Kpn I 酶切位点的遗传多态性以及该位点等位基因片段传递的规律, 采用PCR-RFLP技术, 对162例无血缘关系的健康中国汉人的324条 染色体进行检测,另又对5个家系16位成员进行同样的检测,然后用χ2检验进行统计学处理。结果表明,等位基因A1 频率为0.17,等位基因A2 频率为0.83,杂合率为29%;A1 、A2 的传递规律与理论上预计的完全符合。认为中国汉族群体IDUA 基因 Kpn I 酶切位点也具有遗传多态性, 并且与国外报道的无显著性差异;A1、A2 在世代中的传递完全符合孟德尔遗传规律。
Abstract:To investigate the genetic polymorphism of the Kpn I site in the α-L-iduronidase(IDUA) gene from a Han population in southern China and to study the mode of transmission of alleles, PCR-RFLP was used to analyze 324 chromosomes from 162 Chinese unrelated healthy Han individuals, and the analysis of the genotypes of 16 members in five families. To compare the frequencies and heterzygosity between Chinese Han population and Caucasians in Western by using χ2test. The frequency of allele 1 (450bp) was 0.17,allele 2 (390 plus 60 bp) 0.83, the heterozygosity was 29%.The genotypes of each member of all families detected was completely agreement with the theorical assessment. The locus of Kpn I in the IDUAgene from Han population has polymorphism. There is no significant difference between Chinese Han population and Caucasians in Western countries. The transmission of alleles was agreement with the Mendelian genetic law. 相似文献

Three Asian green pit vipers were collected in August 2013 during a field trip in Fanjin Mt. National Conservation Area, Guizhou. These specimens were identified as Sinovipera sichuanensis, based on subsequent examination and comparison. VRC01,a broadly neutralizing monoclonal antibody(bnmAb),can neutralize a diverse array of HIV-1 isolates by mimicking CD4 binding to the envelope glycoprotein gp120.We have previously demonstrated the presence of VRC01-resistant strains in an HIV-1 infected patient during antiretroviral therapy.Here,we report follow-up studies of two subsequent samples from the same patient.With genetic and phenotypic analysis of over 70 full-length molecular clones of the HIV-1 envelope,we show that VRC01-resistant HIV-1 continued to exist and change in its proportion of the infecting virus during treatment with a highly active antiretroviral therapy.Consistent with our previous observation,the resistant phenotype was associated with a single asparagine residue at position 460(N460),a potential N-linked glycosylation site in the V5 region.The persistence and continuing evolution of VRC01-resistant HIV-1 in vivo presents a great challenge to our future preventative and therapeutic interventions based on VRC01. 相似文献
